Residual solvents: the questions a purity percentage leaves open
How to assess a solvent panel, reporting limit, and lot-specific result without confusing it with HPLC purity.
Manufacturing and purification can involve organic solvents. A high chromatographic purity result does not tell you whether a separate solvent specification was met. Residual-solvent review therefore begins with the manufacturing question: which solvents could reasonably remain, and did the laboratory use a suitable method to look for them?
A separate analytical question
ICH Q3C groups pharmaceutical residual solvents according to their toxicological concern. It is a framework for pharmaceutical quality, not a universal pass label for every research reagent. A research laboratory should define acceptance criteria for its own application with appropriate expertise. Do not convert a pharmaceutical exposure limit into a claim that an unapproved research vial is suitable for administration.
Ask what the panel covers
Request the named solvents, sample preparation, analytical technique, reporting units, and detection or quantitation limits. A statement such as solvent tested is incomplete when the target list is absent. Headspace gas chromatography is one common approach, but the method must fit the sample and target analytes. A result below a reporting limit means below that limit under that procedure, not proven absence of every solvent.
Interpret the result beside its units
Concentration may be expressed as parts per million or a mass fraction. Compare a reported value only with a specification expressed on a compatible basis. Ask whether the result describes the material as received or a dried basis. An unexplained unit conversion can create apparent disagreement between otherwise compatible records. Preserve the original result, then document any calculation used for an internal assessment.
Build the procurement question correctly
Before purchase, list the solvent evidence needed for the intended assay. After receipt, verify that the certificate identifies the same lot and presentation. If only an HPLC trace is available, record solvent status as unreported rather than inferred clean. Unexpected assay behavior may justify additional analytical work, but a failed biological experiment alone does not identify a particular solvent. Keep the chemical measurement and the biological observation as separate findings.
