Endotoxin testing: method, interference, and reporting limits

What an endotoxin result answers, why assay suitability matters, and how it differs from sterility.

Endotoxin is an analytical concern distinct from peptide identity and chromatographic purity. A sample can give a clean-looking chromatogram while lacking any reported endotoxin assessment. When this measurement matters to a study, the question is not whether a seller uses the word tested. It is whether a suitable assay produced a traceable result.

Identify the assay

The FDA's current guidance discusses bacterial endotoxin methods and the relationship to USP chapters, including recombinant-reagent testing. Read the laboratory's actual method rather than assuming all reports use the same reagent system. A certificate should identify the measured property and reporting units. Methods and acceptance criteria designed for pharmaceutical products should not be presented as blanket certification of an unrelated research material.

Check the sample matrix

A sample can interfere with a biological analytical system. Ask whether the lab evaluated inhibition or enhancement, what dilution was used, and whether the reporting limit still answers the study's question after dilution. A highly diluted specimen may become analytically compatible while producing a less informative upper bound. Method suitability and sensitivity need to be considered together, not treated as interchangeable signs of quality.

Separate three different claims

Endotoxin testing addresses a particular class of bacterial material. Sterility testing asks whether viable microorganisms are detected under a defined procedure. A general pyrogen assessment can involve a broader biological question. None of these terms should be substituted for another on a product page. If a report lists only one, mark the others unreported. A sealed cap and a white powder provide no analytical answer to any of them.

Decide what your experiment requires

Cell type, readout, sample matrix, and study design all influence whether an endotoxin result is sufficiently informative. Define requirements before comparing suppliers. Keep the lot, original certificate, method, result, reporting limit, and internal acceptance decision together. When a result is unclear, obtain clarification from the issuing laboratory instead of relabeling it as a pass. The objective is an interpretable measurement, not an impressive badge.
